Induction of Chronic Pneumonia in Normal Mice by Klebsiella pneumoniae
نویسندگان
چکیده
Experimental chronic pneumonia was established in normal BALB/c mice employing a virulent encapsulated strain of Klebsiella pneumoniae B5055. A single exposure to the bacterial pathogen resulted in acute pneumonia that resolved within a week. To achieve the chronic state of lung infection, repeated infection was given via intranasal route. Mice were inoculated with 50ul of 10 4 CFU/ml each on day 0, 7 and 14. There were two peaks observed in the assessment of the lung bacterial load and pro inflammatory cytokine generation following the first and the third infection, respectively. On day 18 of infection, histopathological examination revealed extensive chronic inflammatory changes in the lungs of infected mice which coincided with high lung bacterial load. Tissue damage was also assessed through analysis of malondialdehyde (MDA), lactate dehydrogenase (LDH), and nitric oxide generation in the lung homogenates of the infected mice.

Receptor for Advanced Glycation End Products (RAGE) Serves a Protective Role during Klebsiella pneumoniae - Induced Pneumonia
Klebsiella species is the second most commonly isolated gram-negative organism in sepsis and a frequent causative pathogen in pneumonia. The receptor for advanced glycation end products (RAGE) is expressed on different cell types and plays a key role in diverse inflammatory responses. We here aimed to investigate the role of RAGE in the host response to Klebsiella (K.) pneumoniae pneumonia and ...
متن کاملInduction & resolution of lobar pneumonia following intranasal instillation with Klebsiella pneumoniae in mice.
BACKGROUND & OBJECTIVES Pneumonia caused by Klebsiella pneumoniae is important due to its high morbidity and mortality, especially in context of nosocomial infections. Many experimental studies have focused on the induction and progression of infection till it peaks, but the process of resolution has not been described. In the present study, we successfully attempted to establish an acute respi...
متن کاملDivergent roles of IL-23 and IL-12 in host defense against Klebsiella pneumoniae
Interleukin (IL)-23 is a heterodimeric cytokine that shares the identical p40 subunit as IL-12 but exhibits a unique p19 subunit similar to IL-12 p35. IL-12/23 p40, interferon gamma (IFN-gamma), and IL-17 are critical for host defense against Klebsiella pneumoniae. In vitro, K. pneumoniae-pulsed dendritic cell culture supernatants elicit T cell IL-17 production in a IL-23-dependent manner. Howe...
متن کاملEvaluation of antibiotic resistance associated with ophthalmic oqxAB pumps in Klebsiella pneumoniae causing urinary tract infection
Background: The urinary tract infection is the secondary most common human infection. Colon bacteria, including Klebsiella pneumonia, are the main cause of urinary tract infections. The occurrence of the antibiotic resistance is a major problem in the treatment of infections. Beta- lactamases and efflux pumps constitute the major defense mechanisms of antibiotic resistance of Klebsiella pneumon...
متن کاملAlcohol-associated intestinal dysbiosis impairs pulmonary host defense against Klebsiella pneumoniae
Chronic alcohol consumption perturbs the normal intestinal microbial communities (dysbiosis). To investigate the relationship between alcohol-mediated dysbiosis and pulmonary host defense we developed a fecal adoptive transfer model, which allows us to investigate the impact of alcohol-induced gut dysbiosis on host immune response to an infectious challenge at a distal organ, independent of pre...
